We review recent and anticipated progress on isoform-selective drugs and chemical probes that target Phosphoinositide-3-kinases (PI3Ks). The current lack of isoform-selective, covalent or allosteric PI3K inhibitors limits both our understanding of the complex physiological functions of PI3Ks but also the exploitation of their therapeutic potential.
