Das Oschkinat-Labor konzentriert sich auf die strukturelle Charakterisierung von Protein-Protein-Interaktionen, die für den Empfang und die Weiterleitung von Signalen in biologischen Systemen verantwortlich sind. Wir interessieren uns für Proteindomänen, die spezifische Peptide erkennen, sowie für membranintegrierte Rezeptoren und Rezeptor-Liganden-Komplexe.


Hochkomplexe Strukturinformationen untersuchen

Die Festkörper-NMR-Spektroskopie mit Magic-Angle-Spinning (MAS) ist in der Lage, hochauflösende Strukturinformationen von komplexen Proben biologischer Makromoleküle zu liefern, unabhängig von deren Molekulargewicht. Die MAS-NMR-Spektroskopie ist eine attraktive Methode für strukturelle Untersuchungen an kleinen Proteinen, die in Lipiddoppelschichten eingebettet sind, großen polydispersen Komplexen oder Proteinen in ihrer natürlichen Umgebung, beispielsweise in lebenden Biofilmen. Als besondere Vorteile kann NMR die Position von Protonen sowie ihren chemischen Austausch erfassen und schnell interkonvertierende Zustände auf der Basis ihrer charakteristischen chemischen Verschiebungen unterscheiden. Da die MAS-NMR-Spektroskopie die Untersuchung heterogener Proben ermöglicht, wollen wir Strukturuntersuchungen im “realen Raum” einer Zelle durchführen, wobei wir langfristig von einer 20-100-fachen Erhöhung des Signal-Rausch-Verhältnisses durch den Einsatz der dynamischen Kernpolarisation (DNP) profitieren. Zu diesem Zweck haben wir DNP-Methoden an biologischen Proben verbessert und wenden eine sehr schnelle Rotation in der MAS-NMR (100.000 Rotationen pro Sekunde) zur Untersuchung biologischer Proben an. Bei hohen MAS-Geschwindigkeiten können mit einer minimalen Probenmenge hochauflösende Protonenspektren erhalten werden. Schnelle MAS bei 100 kHz wurde beispielsweise zur Untersuchung der Bindung kleiner Moleküle an den neonatalen Fc-Rezeptor verwendet. Desweiteren untersuchen wir große dynamische und polydisperse Proteinsysteme, die an der Proteinhomöostase beteiligt sind, einschließlich kleiner Hitzeschockproteine, biologische Systeme die in Phasentrennung involviert sind sowie Biofilme.

 

Research Focus

The main focus of our group is the development of solid-state magic-angle spinning (MAS) NMR methodology as a routine tool for biological studies, in particular for structural characterisation of protein-protein interactions that are responsible for the reception and transduction of signals. This research focuses traditionally on membrane integrated proteins and receptor-ligand complexes.

In recent years a new method for signal enhancement called dynamic nuclear polarisation (DNP) was established and its value tested in structure determination and metabolomic projects. Using DNP, the nascent peptide chain in the tunnel of the ribosome was investigated, and the nature of black deposits in cartilage of Alkaptonuria patients, who suffer from impaired tyrosine degradation. Recently, we started a set of new projects on membrane proteins and inositol lipids in their actual membranes, and on live biofilms. Furthermore, we have started a project on the principles underlying liquid-liquid phase separation, with the protein FUS as an example. Our aim is to provide quantitative information as to individual interactions, responsible for establishing the dense phase and the structural characterization of such interactions. Furthermore, we are investigating proton flows and proton dynamics to understand protein function, funded by the SFB 1078.
